摘要

目的 建立12批火把花根片的超高效液相色谱(UPLC)指纹图谱,评价其批间质量一致性,并对主要化学成分进行鉴定,以明确其物质基础。方法 采用Waters CORTECS T3色谱柱(2.1 mm×100 mm,1.6 μm),乙腈-0.1%甲酸水溶液为流动相,梯度洗脱,流速为0.4 mL·min-1,检测波长为260 nm,柱温为35 ℃。利用“中药色谱指纹图谱相似度评价系统(2012版)”进行相似度分析。采用超高效液相色谱四极杆串联飞行时间质谱(UPLC-Q-TOF-MS)技术对火把花根片的主要成分进行鉴定。结果 建立的火把花根片UPLC指纹图谱共标定了27个共有峰,12批样品的相似度为0.992~0.999,鉴定了18个主要成分,多数为黄酮和酚苷类成分。结论 火把花根片批间质量一致性较好,所含成分的极性相对较大,与该制剂水提醇沉的生产工艺相关。本研究中建立的指纹图谱和主要成分的结构鉴定可为后期该品种的质量控制研究提供依据。

Abstract

OBJECTIVE To evaluate the inter-batch quality consistency of 12 batches of Huobahuagen tablets using UPLC fingerprints, and to identify the main chemical constituents for clarifying their substance bases. METHODS Chromatographic separation was performed on a Waters CORTECS T3 column (2.1 mm×100 mm, 1.6 μm). Acetonitrile-0.1% formic acid aqueous solution was adopted as mobile phase for gradient elution. The flow rate was set at 0.4 mL·min-1. The detection wavelength was set at 260 nm. The column temperature was maintained at 35 ℃. The similarity of fingerprint was evaluated by the “Similarity Evaluatio System of Chromatographic Fingerprint of Traditional Chinese Medicine” (2012 edition). At the same time, the main chemical constituents of Huobahuagen tablets were identified by UPLC-Q-TOF-MS method. RESULTS In the established fingerprint, a total of 27 common peaks were calibrated. The similarity of the tested samples was 0.992-0.999. Eighteen main constituents were identified, most of which were phenolic glycosides and flavonoids. CONCLUSION Huobahuagen tablets have good inter-batch quality consistency. The main components contained in this preparation have relatively high polarity, which is due to the production process of water extraction and alcohol precipitation. The results presented above can serve as a foundation for a later quality control study of this preparation.
